The conventional paradigm for developing new treatments for disease mainly involves either the discovery of new drug targets, or finding new, improved drugs for old targets. However, an ion channel found only in invertebrates offers the potential of a completely new paradigm in which an established drug target can be re‐engineered to serve as a new candidate therapeutic agent. The L‐glutamate‐gated chloride channels (GluCls) of invertebrates are absent from vertebrate genomes, offering the opportunity to introduce this exogenous, inhibitory, L‐glutamate receptor (...) into vertebrate neuronal circuits either as a tool with which to study neural networks, or a candidate therapy. Epileptic seizures can involve L‐glutamate‐induced hyper‐excitation and toxicity. Variant GluCls, with their inhibitory responses to L‐glutamate, when engineered into human neurons, might counter the excitotoxic effects of excess L‐glutamate. In reviewing recent studies on model organisms, it appears that this approach might offer a new paradigm for the development of candidate therapeutics for epilepsy. (shrink)

Suicide among Veterans experiencing or at risk for homelessness remains a significant public health concern. Conducting research to understand and meet the needs of this at-risk population remains challenging due to myriad factors. To address this challenge, the United States Department of Veterans Affairs convened the Health Services Research and Development Suicide Prevention in Veterans Experiencing Homelessness: Research and Practice Development meeting, bringing together subject-matter experts in the fields of homelessness and suicide prevention, both from within and outside of VA. (...) During the meeting, attendees identified 10 potential research priorities at the intersection of suicide prevention and homelessness. After the meeting, Delphi methodology was used to achieve consensus on the relative importance of the identified research domains. Through this iterative Delphi process, agreement was reached regarding the need to increase understanding of barriers and facilitators to suicide risk assessment and emergency intervention for Veterans experiencing homelessness by examining the perspectives of both Veterans and healthcare providers. Elucidating the complex relationships between risk periods, subgroups, suicide means, and drivers of suicide among Veterans experiencing homelessness was also considered a top priority. This article documents the Delphi process and provides a research agenda for researchers, funding agencies, and policymakers to prioritize the most relevant and potentially impactful research domains aimed at preventing suicide among Veterans experiencing or at risk for homelessness. DBS Think Tank IX was held on August 25–27, 2021 in Orlando FL with US based participants largely in person and overseas participants joining by video conferencing technology. The DBS Think Tank was founded in 2012 and provides an open platform where clinicians, engineers and researchers can freely discuss current and emerging deep brain stimulation technologies as well as the logistical and ethical issues facing the field. The consensus among the DBS Think Tank IX speakers was that DBS expanded in (...) its scope and has been applied to multiple brain disorders in an effort to modulate neural circuitry. After collectively sharing our experiences, it was estimated that globally more than 230,000 DBS devices have been implanted for neurological and neuropsychiatric disorders. As such, this year’s meeting was focused on advances in the following areas: neuromodulation in Europe, Asia and Australia; cutting-edge technologies, neuroethics, interventional psychiatry, adaptive DBS, neuromodulation for pain, network neuromodulation for epilepsy and neuromodulation for traumatic brain injury. Based on brain imaging findings, we present a model according to which addiction emerges as an imbalance in the information processing and integration among various brain circuits and functions. The dysfunctions reflect (a) decreased sensitivity of reward circuits, (b) enhanced sensitivity of memory circuits to conditioned expectations to drugs and drug cues, stress reactivity, and (c) negative mood, and a weakened control circuit. Although initial experimentation with a drug of abuse is largely a voluntary behavior, continued drug use can eventually (...) impair neuronal circuits in the brain that are involved in free will, turning drug use into an automatic compulsive behavior. The ability of addictive drugs to co‐opt neurotransmitter signals between neurons (including dopamine, glutamate, and GABA) modifies the function of different neuronal circuits, which begin to falter at different stages of an addiction trajectory. Upon exposure to the drug, drug cues or stress this results in unrestrained hyperactivation of the motivation/drive circuit that results in the compulsive drug intake that characterizes addiction. ‘Without exaggeration and oversimplification little progress is made in most fields of humanistic investigation.’ With this disarming quotation from A. D. Nock, Albert Henrichs begins his book-length interpretation of P. Colon, inv. 3328. In the same spirit of humanistic progress, I would like to reconsider some aspects of the text and to offer a different assessment of its place in the history of religion and literature.The fragments are from three pages of a hitherto unknown Greek novel, Lollianos'Phoinikika. Frags A and (...) B luckily include book-ends, from whose subscriptions we know the author and title of the work. Frag. C is just scraps which yield no continuous sense. Frag. A brokenly and confusedly mentions youths, women dancing, being thrown off the roof, sobriety, kissing, and then, in a slightly more intelligible scene, the male narrator's loss of virginity with a woman named Persis, her gift to him of a gold necklace which he refuses, the assistance of one Glauketes in taking the necklace elsewhere, and finally what seems to be a confrontation between Persis' mother and the two lovers. This last is similar to Achilles Tatius ii 23–5. Achilles Tatius also offers the closest parallel to frag. B, a ghastly description of human sacrifice and cannibalism. This scene is the focus of most of Henrichs' interpretation and I will limit myself to it in the present article.The central question raised by this new novel fragment is how to assess the relative importance of religious and literary parallels. Is thePhoinikikato be regarded as a document in religious or in literary history, or perhaps somewhere on the borderland of both? There has been a lively discussion in the last half century of the thesis that the ancient novels were written and read as religious documents, deriving their basic structure and many details from the myths and cults of particular religions. Henrichs devotes most of his book to arguing that the sacrifice scene in Lollianos is inspired by an actual rite, probably of a Dionysian character, and that thePhoinikikaserves to illuminate a little-known corner of religious history. His views are based on an extensive collection of liturgical, mythical and ethnological parallels concerning oath rituals, the sacrifice of children, cannibalism, and face-painting. Of all the parallels cited, the two which are closest in every way to Lollianos are Achilles Tatius iii 15 and Cassius Dio lxxi 4. On the strength of these Henrichs asserts that Lollianos' description of a ritual murder represents, more or less directly, the cultic practice of the Egyptian Boukoloi. Without postulating a religious message for thePhoinikikaas a whole, Henrichs does claim that this scene yields valuable information about the structure of ancient mystery rituals and that these new fragments support the methodological correctness of Kerenyi's and Merkelbach's approach to the ancient novels. Copeland goes on to analyze what those working in AI must achieve before they can claim to have built a thinking machine and appraises their prospects of succeeding.There are clear introductions to connectionism and to the language of thought hypothesis which weave together material from philosophy, artificial intelligence and neuroscience. Turing''s test has been much misunderstood. Recently unpublished material by Turing casts fresh light on his thinking and dispels a number of philosophical myths concerning the Turing test. Properly understood, the Turing test withstands objections that are popularly believed to be fatal.

To compute is to execute an algorithm. More precisely, to say that a device or organ computes is to say that there exists a modelling relationship of a certain kind between it and a formal specification of an algorithm and supporting architecture. The key issue is to delimit the phrase of a certain kind. I call this the problem of distinguishing between standard and nonstandard models of computation. The successful drawing of this distinction guards Turing's 1936 analysis of computation against (...) a difficulty that has persistently been raised against it, and undercuts various objections that have been made to the computational theory of mind. Accelerating Turing machines are Turing machines of a sort able to perform tasks that are commonly regarded as impossible for Turing machines. For example, they can determine whether or not the decimal representation of contains n consecutive 7s, for any n; solve the Turing-machine halting problem; and decide the predicate calculus. Are accelerating Turing machines, then, logically impossible devices? I argue that they are not. There are implications concerning the nature of effective procedures and the theoretical limits of computability. Contrary (...) to a recent paper by Bringsjord, Bello and Ferrucci, however, the concept of an accelerating Turing machine cannot be used to shove up Searle's Chinese room argument. (shrink)

Accelerating Turing machines have attracted much attention in the last decade or so. They have been described as “the work-horse of hypercomputation” (Potgieter and Rosinger 2010: 853). But do they really compute beyond the “Turing limit”—e.g., compute the halting function? We argue that the answer depends on what you mean by an accelerating Turing machine, on what you mean by computation, and even on what you mean by a Turing machine. We show first that in the current literature the term (...) “accelerating Turing machine” is used to refer to two very different species of accelerating machine, which we call end-stage-in and end-stage-out machines, respectively. We argue that end-stage-in accelerating machines are not Turing machines at all. We then present two differing conceptions of computation, the internal and the external, and introduce the notion of an epistemic embedding of a computation. We argue that no accelerating Turing machine computes the halting function in the internal sense. Finally, we distinguish between two very different conceptions of the Turing machine, the purist conception and the realist conception; and we argue that Turing himself was no subscriber to the purist conception. We conclude that under the realist conception, but not under the purist conception, an accelerating Turing machine is able to compute the halting function in the external sense. We adopt a relatively informal approach throughout, since we take the key issues to be philosophical rather than mathematical. (shrink)

This paper charts some early history of the possible worlds semantics for modal logic, starting with the pioneering work of Prior and Meredith. The contributions of Geach, Hintikka, Kanger, Kripke, Montague, and Smiley are also discussed.

The revolution in semantics in the late 1960s and 1970s overturned an earlier competing paradigm, ‘translational’ semantics. I revive and defend Prior’s translational semantics for modals and tense-modals. I also show how to extend Prior’s propositional modal semantics to quantificational modal logic, and use the resulting semantics to formalize Prior’s own counterexample to the Barcan Formula.

What are the limits of physical computation? In his ‘Church’s Thesis and Principles for Mechanisms’, Turing’s student Robin Gandy proved that any machine satisfying four idealised physical ‘principles’ is equivalent to some Turing machine. Gandy’s four principles in effect define a class of computing machines (‘Gandy machines’). Our question is: What is the relationship of this class to the class of all (ideal) physical computing machines? Gandy himself suggests that the relationship is identity. We do not share this view. The mathematical genius Alan Turing, well known for his crucial wartime role in breaking the ENIGMA code, was the first to conceive of the fundamental principle of the modern computer. This text contains first hand accounts by Turing and by the pioneers of computing who worked with him on his revolutionary design for an electronic computing machine - his Automatic Computing Engine.

Respected modern scholars regard the pre-Buddhist philosopher Sañjaya Belaṭṭhiputta—a significant figure in the Buddhist canon—as the originator of the important classical argument- forms known as the catuṣkoṭi and catuṣkoṭi vinirmukta. We argue that the early Buddhist texts do not in fact support this view of the origin of these argument-forms; the question of their origin is open. While it is certainly true that the Pāli Sāmaññaphala Sutta and some of its parallels portray Sañjaya as deploying the catuṣkoṭi, there is nothing (...) in these passages to suggest that he was its originator. The situation concerning the catuṣkoṭi vinirmukta is perhaps even more surprising: There is nothing in the early Pāli texts and their parallels—nor in Buddhagosa’s famous Pāli commentary on the early texts—to show that Sañjaya even deployed the catuṣkoṭi vinirmukta let alone originated it. Further, our investigations also call into question the standard portrayal of Sañjaya as an obfuscator and prevaricator. It appears he may have been a more interesting and able philosopher than the Sāmaññaphala Sutta—and modern accounts based on it—maintain. (shrink)
